Piperlongumine has anti-cancer activity in numerous cancer cell lines via various signaling pathways. But there been no study regarding the mechanisms of PL on lung yet. Thus, we evaluated effects and possible non-small (NSCLC) cells vivo vitro. Our findings showed that induced apoptotic death suppressed DNA binding NF-κB a concentration dependent manner (0-15 μM) NSCLC cells. Docking model pull down assay directly binds to site nuclear factor-κB (NF-κB) p50 subunit, surface plasmon...

Transforming growth factor-beta (TGF-β), a pluripotent cytokine expressed in the colon, has crucial but paradoxical role colorectal cancer (CRC). TGF-β is potent proliferation inhibitor of normal colon epithelial cells and acts as tumor suppressor. However, also promotes invasion metastasis during late-stage CRC, thereby acting an oncogene. Thus, understanding factors behind roles elucidating mechanisms by which TGF-β-induced inhibition impaired CRC are necessary. Here, we found that N-Myc...

Abstract Acquired resistance to osimertinib (Osi) remains a major obstacle in the treatment of patients with EGFR‐mutant non‐small cell lung cancer (NSCLC). AXL elevation is known key mechanism Osi‐resistance, and therapeutic strategies remain scarce. Emerging evidence reveals that an increased intracellular glutathione (GSH) level induces Osi resistance. In this study, new identified by which GSH regulates expression via peroxidase 4 (GPX4) Osi‐resistant cells. A multifunctional covalent...

Background Despite advancements in treatment modalities, several patients with colorectal cancer (CRC) remain unresponsive to immune checkpoint inhibitor therapy. Pyroptosis, an inflammatory programmed cell death process, holds substantial promise for tumor immunotherapy. In this study, we explored the use of pyroptosis overcome immunotherapy resistance CRC. Methods We used a pyroptosis-related gene panel construct efficacy evaluation model and validated its performance by...

Background Cervical lymph node metastasis is a major factor influencing recurrence after surgery for papillary thyroid cancer. Molecular markers that can predict the presence of and assess aggressiveness microcarcinoma (PTMC) remain poorly understood. The research question addressed whether specific genes, such as thrombospondin-4 (THBS4), could serve predictive biomarkers guiding surgical strategies, particularly in cases where current imaging modalities fail to detect LNM central region,...

Rationale: Signal transducer and activator of transcription-3 (STAT3) plays a pivotal role in cancer biology. Many small-molecule inhibitors that target STAT3 have been developed as potential anticancer drugs. While designing the SH2 domain remains leading focus for drug discovery, there has growing interest targeting DNA-binding (DBD) protein. Methods: We demonstrated antitumor activity novel, (E)-2-methoxy-4-(3-(4-methoxyphenyl)prop-1-en-1-yl)phenol (MMPP) directly binds to DBD STAT3,...

Signal transducer and activator of transcription 3 (STAT3) nuclear factor-κB (NF-κB) are two representative factors that play a critical role in inflammation-associated tumorigenesis through multi-level cooperation. Unlike other types tumors, breast carcinomas have shown significant dependency on the non-classical NF-κB pathway as well classical one. The α subunit inhibitor κB kinase (IKK) complex, IKKα, is involved both activation NF-κB. Although cross-talk between STAT3 has been suggested...

In the present study, we synthesized several non‐aldehyde analogues of (E)‐2,4‐bis(p‐hydroxyphenyl)‐2‐butenal which showed anti‐cancer effect. Interestingly, among 16 compounds, found that (E)‐2‐methoxy‐4‐(3‐(4‐methoxyphenyl)prop‐1‐en‐1‐yl)phenol (MMPP) most significant anti‐proliferative effect on PA‐1 and SK‐OV‐3 ovarian epithelial cancer cells.
