Abstract BRD4, a Bromodomain and Extraterminal (BET) protein family member, is promising anti-cancer drug target. However, resistance to BET inhibitors targeting BRD4 common in solid tumors. Here, we show that cancer-associated fibroblast (CAF)-activated stromal signaling, interleukin-6/8-JAK2, induces phosphorylation at tyrosine 97/98 colorectal cancer, resulting stabilization due interaction with the deubiquitinase UCHL3. also displays increased binding chromatin but reduced inhibitors,...

Abstract Dysregulation of the cell cycle machinery leads to genomic instability and is a hallmark cancer associated with chemoresistance poor prognosis in colorectal (CRC). Identifying targeting aberrant expected improve current therapies for CRC patients. Here, upregulated polo‐like kinase 1 (PLK1) signaling, accompanied by deregulation cycle‐related pathways identified. It shown that PLK1 signaling correlates recurrence Genetic pharmacological blockade significantly increases sensitivity...

Abstract Cancer-associated fibroblasts (CAFs) have been shown to play a strong role in colorectal cancer metastasis, yet the underlying mechanism remains be fully elucidated. Using CRC clinical samples together with ex vivo CAFs-CRC co-culture models, we found that CAFs induce expression of Leucine Rich Alpha-2-Glycoprotein 1(LRG1) CRC, where it shows markedly higher metastatic tissues compared primary tumors. We further show CAFs-induced LRG1 promotes migration and invasion is concomitant...

Increasing interest has developed in the therapeutic potential of bone marrow-derived mesenchymal stem cells (MSCs) for treatment inflammatory bowel disease (IBD) and IBD-induced cancer. However, whether MSCs have ability to suppress or promote tumor development remains controversial. The stromal cell-derived factor 1 (SDF-1)/C-X-C chemokine receptor type 4 (CXCR4) axis is well known play a critical role homing MSCs. In this study, we aimed evaluate CXCR4-overexpressing on tumorigenesis IBD....

Pancreatic adenocarcinoma (PAAD) is a treatment-refractory cancer with poor prognosis. Accumulating evidence suggests that squalene epoxidase (SQLE) plays pivotal role in the development and progression of several types humans. However, function underlying mechanism SQLE PAAD remain unclear.SQLE expression data were downloaded from The Cancer Genome Atlas Genotype-Tissue Expression database. alterations demonstrated based on cBioPortal upstream miRNAs regulating predicted using starBase....

Resistance to neoadjuvant chemotherapy leads poor prognosis of locally advanced rectal cancer (LARC), representing an unmet clinical need that demands further exploration therapeutic strategies improve outcomes. Here, we identified a noncanonical role RB1 for modulating chromatin activity contributes oxaliplatin resistance in colorectal (CRC). We demonstrate induces phosphorylation, which is associated with the oxaliplatin-based LARC. Inhibition phosphorylation by CDK4/6 inhibitor results...

Abstract In‐stent restenosis caused by tumor ingrowth is a major problem for patients undergoing stent placement because conventional stents often lack sustainable antitumor capabilities. The aim of this work to develop silk fibroin (SF)‐based nanofibrous membrane that loaded with combined‐therapy drugs using electrospinning technologies, which further coated on polydioxanone (PDO) and used the treatment colorectal cancer (CRC). In order improve effectiveness, combination therapeutic drugs,...

Abstract Despite showing promise against PIK3CA-mutant breast cancers in preclinical studies, PI3K/AKT pathway inhibitors demonstrate limited clinical efficacy as monotherapy. Here, we found that histone H3K27me3 demethylase KDM6B-targeted IGFBP5 expression provides a protective mechanism for inhibitor-induced apoptosis cancer cells. We overexpression of KDM6B and luminal are positively associated with poorer disease outcomes. Mechanistically, promotes by antagonizing EZH2-mediated...

EZH2 is over-expressed in human colon cancer and closely associated with tumor proliferation, metastasis poor prognosis. Targeting inhibiting may be an effective therapeutic strategy for cancer. 3-Deazaneplanocin A (DZNep), as inhibitor, can suppress cell growth. However, the anti-cancer role of DZNep cells has been rarely studied. In this study, we demonstrate that inhibit growth survival HCT116 by inducing cellular senescence apoptosis.
