Enhancer of Zeste homolog 2 (EZH2) is a methyltransferase that plays an important role in many biological processes through its ability to trimethylate lysine 27 histone H3. Here, we show Akt phosphorylates EZH2 at serine 21 and suppresses activity by impeding binding H3, which results decrease trimethylation derepression silenced genes. Our imply regulates the methylation activity, phosphorylation EZH2, may contribute oncogenesis.
Breast cancers are highly heterogeneous but can be grouped into subtypes based on several criteria, including level of expression certain markers. Claudin-low breast cancer (CLBC) is associated with early metastasis and resistance to chemotherapy, while gene profiling indicates it characterized by the markers epithelial-mesenchymal transition (EMT) - a phenotypic conversion linked metastasis. Although epigenetic program controlling cellular plasticity EMT remains unclear, one contributor may...
Snail, a key inducer of epithelial-mesenchymal transition (EMT), plays an important role in cancer metastasis. To better understand the Snail metastasis ovarian carcinoma, expression was knocked down by antisense-Snail highly metastatic cell line HO8910PM. Gene array analysis revealed that blocking suppressed activity matrix metalloproteinases (MMPs) and upregulated TIMP3, MMP inhibitor. These findings suggest interacts with during tumor invasion In addition, we examined orthotopic model...
Abstract Snail1, a key transcription factor of epithelial–mesenchymal transition (EMT), is subjected to ubiquitination and degradation, but the mechanism by which Snail1 stabilized in tumours remains unclear. We identify Dub3 as bona fide deubiquitinase, interacts with stabilizes Snail1. overexpressed breast cancer; knockdown resulted destabilization, suppressed EMT decreased tumour cell migration, invasion, metastasis. These effects are rescued ectopic expression. IL-6 also inducing...

Abstract BRD4 assembles transcriptional machinery at gene super-enhancer regions and governs the expression of genes that are critical for cancer progression. However, it remains unclear whether BRD4-mediated transcription is required tumor cells to develop drug resistance. Our data show prolonged treatment luminal breast with AKT inhibitors induces FOXO3a dephosphorylation, nuclear translocation, disrupts its association SirT6, eventually leading acetylation as well recognition. Acetylated...

Cyclin-dependent kinase 9 (CDK9) plays a critical role in transcription initiation and is essential for maintaining gene silencing at heterochromatic loci. Inhibition of CDK9 increases sensitivity to immunotherapy, but the underlying mechanism remains unclear. We now report that RNF20 stabilizes LSD1 via K29-mediated ubiquitination, which dependent on CDK9-mediated phosphorylation. This CDK9- RNF20-dependent stabilization necessary demethylation histone H3K4, then subsequent repression...
Previous studies have indicated that inflammatory stimulation represses protein phosphatase 2A (PP2A), a well-known tumor suppressor. However, whether PP2A repression participates in pancreatic cancer progression has not been verified. We used lipopolysaccharide (LPS) and macrophage-conditioned medium (MCM) to establish vitro inflammation models, investigated stimuli affect cell growth invasion catalytic subunit (PP2Ac)-dependently. Via nude mouse models of orthotopic xenografts dibutyltin...
Thrombospondin 1 (TSP1) is a multifunctional matricellular protein. Recent studies demonstrate that TSP1 highly expressed in adipose tissue (AT) and positively associated with AT inflammation insulin resistance (IR). In this study, the contribution of different cellular sources to obesity-induced metabolic complications determined by using mice either adipocyte or myeloid/macrophage-specific deletion diet-induced obese model. The results demonstrated neither nor affected development...
To evaluate the performance of a composite scaffold Wharton's jelly (WJ) and chondroitin sulfate (CS) effect loaded with human umbilical cord mesenchymal stem cells (hUCMSCs) in repairing articular cartilage defects, two experiments were carried out. The vitro involved identification hUCMSCs, construction biomimetic scaffolds by physical chemical crosslinking WJ CS, testing biomechanical properties both scaffold. In vivo experiments, hUCMSCs applied to repair defects rat knee. Moreover,...
SNAI1 is widely regarded as a master driver of epithelial-mesenchymal transition (EMT) and associated with breast cancer progression metastasis. This pro-malignant role strongly linked to posttranslational modification, especially phosphorylation, which controls its protein levels subcellular localization. While multiple kinases are implicated in regulation stability, the precise mechanism by stabilized tumors remains be fully elucidated. Methods: A series vitro vivo experiments were...
Tetraspanin CD151 interacts with laminin-binding integrins (i.e., α3β1, α6β1 and α6β4) other cell surface molecules to control diverse cellular physiological processes, ranging from adhesion, migration survival tissue architecture homeostasis. Here, we report a novel role of in maintaining the branching morphogenesis activity progenitor cells during pubertal development mammary glands. In contrast disruption integrins, removal mice enhanced tertiary glands by 2.4-fold number terminal end...
