Healthy Kupffer cell (KC) pool is dominated by embryonic KCs (EmKCs), preserving liver homeostasis. How the KC varies upon injury remains unclear. Using chimeric mice with bone marrow (BM) cells labeled enhanced green fluorescent protein, we identify that BM monocyte-derived (MoKCs) become dominant in cholestatic- or toxic-injured livers via immunofluorescence and mass cytometry. Single-cell RNA sequencing (scRNA-seq) unveils proliferative, anti-apoptotic properties repair potential of MoKCs...

Macrophage M1/M2 polarization mediates tissue damage and inflammatory responses. Cannabinoid receptor (CB) 1 participated in liver fibrogenesis by affecting bone marrow (BM)-derived monocytes/macrophages (BMMs) activation. However, the knowledge of whether CB1 is involved BMMs remains limited. Here, we found M1 gene signatures (including CD86, MIP-1β, TNF, IL-6 iNOS) amount macrophages (CD86+ cells, gated F4/80) were significantly elevated carbon tetrachloride (CCl4) -induced mouse injured...

Abstract Biliary atresia (BA) is a severe inflammatory and fibrosing neonatal cholangiopathy disease characterized by progressive obstruction of extrahepatic bile ducts, resulting in cholestasis hepatic failure. Cholestasis may play an important role the fibrotic pathological processes, but its specific mechanism still unclear. Necroptosis mediated Z-DNA-binding protein 1 (ZBP1)/phosphorylated-mixed lineage kinase domain-like pseudokinase (p-MLKL) prominent pathogenic factor diseases,...

The major histocompatibility complex (MHC) is one of the most variable and gene-dense regions human genome. Most studies MHC, associated regions, focus on minor variants HLA typing, many which have been demonstrated to be with disease susceptibility metabolic pathways. However, detection in MHC region, diagnostic still lacks a coherent, standardized, cost effective high coverage protocol clinical quality reliability. In this paper, we presented such method for accurate types using...

Using a noninvasive, electro-optically based electron bunch arrival time measurement at FLASH (free laser in Hamburg) the temporal resolution of two-color pump-probe experiments has been significantly improved. The system determines relative extended ultraviolet pulse and an amplified Ti:sapphire femtosecond-laser interaction region better than 90 fs rms. In benchmarking experiment using above threshold ionization noble gases, enhancement timing by factor 4 compared to uncorrected data is obtained.
