Continuous BRAF inhibition of mutant melanomas triggers a series cell state changes that lead to therapy resistance and escape from immune control before establishing acquired genetically. We used genome-wide transcriptomics single-cell phenotyping explore the response kinetics for panel patient-derived BRAFV600 -mutant melanoma lines. A subset plastic lines, which followed trajectory covering multiple known transitions, provided models more detailed biophysical investigations. Markov...

Significance Obtaining cultures of microbes is essential for developing knowledge bacterial genetics and physiology, but many with potential biomedical significance identified from metagenomic studies have not yet been cultured due to the difficulty identifying growth conditions, isolation, characterization. We developed a microfluidics-based, genetically targeted approach address these challenges. This corrects sampling bias differential kinetics, enables use stimulants available only in...

We report the first 7,755 patients with confirmed COVID-19 in Korea as of March 12th, 2020. A total 66 deaths have been recorded, giving a case fatality proportion 0.9%. Older people, and those comorbidities were at higher risk fatal outcome. The highest number cases Daegu, followed by Gyeongbuk. This summary may help to understand disease dynamics early phase outbreaks, therefore, guide future public health measures.

Phosphatidylinositol 3-kinase (PI3K) signaling is highly active in glioblastomas. We assessed pharmacokinetics, pharmacodynamics, and efficacy of the pan-PI3K inhibitor buparlisib patients with recurrent glioblastoma PI3K pathway activation. This study was a multicenter, open-label, multi-arm, phase II trial pathway-activated at first or second recurrence. In cohort 1, scheduled for re-operation after progression received 7 to 13 days before surgery evaluate brain penetration modulation...

We describe chemical approaches for integrated metabolic and proteomic assays from single cells. Quantitative intracellular metabolites, including glucose uptake three other species, are designed as surface-competitive binding with fluorescence readouts. This enables integration into a microarray format functional protein immunoassays, all of which incorporated the microchambers single-cell barcode chip (SCBC). By using SCBC, we interrogate response human-derived glioblastoma cancer cells to...

Malignant pleural effusion (MPE), the presence of malignant cells in fluid, is often first sign many cancers and occurs patients with metastatic malignancies. Accurate detection tumor fluid crucial because MPE denotes an advanced stage disease directs a switch clinical managements. Cytology, as traditional diagnostic tool, has limited sensitivity especially when are not abundant, may be confounded by reactive mesothelial fluid. We describe highly sensitive approach for rapid metabolically...

Chemical investigation of a halophilic actinomycete strain belonging to the genus Nocardiopsis inhabiting hypersaline saltern led discovery new 18-membered macrolides with nitrile functionality, borrelidins C–E (1–3), along previously reported borrelidin (4). The planar structures C–E, which are members rare class antibiotics, were elucidated by NMR, mass, IR, and UV spectroscopic analyses. configurations borrelidines determined interpretation ROESY NMR spectra, J-based configuration...

DNA can be used to store digital data, and synthetic short-sequence pools are developed high quantities of data. However, data cannot actively processed in pools. An active editing process is using splint ligation a droplet-controlled fluidics (DCF) system. fragments discrete sizes (100-500 bps) synthesized for droplet assembly, programmed sequence information exchange occurred. The encoded sequences series parallel synthesize the determined pools, enabling random access polymerase chain...

Amphiphilic discotic molecules with hydrophilic side branches consisting of hexaphenyl hexa-peri-hexabenzocoronene and hexabiphenyl hexa-peri-hexabiphenylcoronene as the aromatic core hexa-substituted oligoethers branched peripheral chains have been synthesized, their microstructure has characterized. The based on dibranched oligoether observed to self-organize into a well-ordered hexagonal columnar structure within liquid crystalline phases, which possessed an exceptionally high thermal...
