Interleukin (IL)-10 is considered a prototypical anti-inflammatory cytokine, significantly contributing to the maintenance and reestablishment of immune homeostasis. Accordingly, it has been shown in intestine that IL-10 produced by Tregs can act on effector T cells, thereby limiting inflammation. Herein, we investigate whether this role also applies cells during central nervous system (CNS) During neuroinflammation, both CNS-resident -infiltrating produce IL-10; yet, as pleotropic function,...

We have cloned a human macrophage receptor that binds to apolipoprotein (apo)B48 of dietary triglyceride (TG)-rich lipoproteins. TG-rich lipoprotein uptake by the apoB48R rapidly converts macrophages and apoB48R-transfected Chinese hamster ovary cells in vitro into lipid-filled foam cells, as seen atherosclerotic lesions. The cDNA (3,744 bp) encodes protein with no known homologs. Its ≈3.8-kb mRNA is expressed primarily reticuloendothelial cells: monocytes, macrophages, endothelial cells....

Background Th17 cells are central pathogenic mediators of autoimmune disease, including many forms GN. IL-10 receptor signaling (IL-10R) in regulatory T (Tregs) has been implicated the downregulation cells, but underlying molecular mechanisms and functional relevance this process remain unclear. Methods We generated mice with Treg-specific IL-10Ra deficiency subjected these to nephrotoxic serum–induced nephritis as a model crescentic Immune responses Treg phenotypes were extensively...

Interleukin 10 (IL-10) is an antiinflammatory cytokine, but also promotes B cell responses and plays a pathogenic role in systemic lupus erythematosus (SLE). CD4 + CCR6 IL-7R T cells from human tonsils produced IL-10 following stimulation by naïve cells, which promoted immunoglobulin G (IgG) production. These tonsillar helper were phenotypically distinct follicular (T FH ) lacked BCL6 expression. In peripheral blood, population with similar characteristics was identified, Th17- -associated...

Lymphocytes have dichotomous functions in ischemic stroke. Regulatory T cells are protective, while IL-17A from innate lymphocytes promotes the infarct growth. With recent advances of cell-subtype specific transgenic mouse models it now has become possible to study complex interplay cell subpopulations stroke. In a murine model experimental stroke we analyzed effects IL-10 on functional outcome for up 14 days post-ischemia and defined source brains based immunohistochemistry, flow cytometry,...

Acidic mammalian chitinase (AMCase) is produced during and plays an important role in the pathogenesis of Th2-mediated diseases antiparasite responses. However, effector responses AMCase these settings have not been adequately defined relationship(s) between its chitinolytic other biologic properties investigated. In studies, we demonstrate that protects airway epithelial cells from Fas ligand- growth factor withdrawal-induced apoptosis. This cytoprotection was associated with Akt...

Abstract Stimulation of T cells through the cell receptor (TcR) initiate activation pathways, and paradoxically can also result in activation‐induced death. Many factors influence a stimulated cell's decision to manifest one or other. Here we show that co‐stimulation with LFA‐1 plays key role choice between two fates, differentiating αβ γδ cells. Peripheral but not undergo apoptosis upon co‐cross‐linking TcR MRL lpr/lpr mice as well +/+ mice. Our results suggest is inducible by combined...

Abstract: A polymorphism in high‐affinity receptor of TNF (TNFR2) gene, Met196Arg, was reported to be associated with systemic lupus erythematosus (SLE) Japanese, whereas the association could not found Europeans at all and this represents an apparent discrepancy. The association, then, should tested other populations clarify possible involvement, if any, TNFR2 SLE or related autoimmune diseases. purposes study were examine Japanese patients investigate its diseases accompanied by...

Background: IL-10 plays a key role in systemic lupus erythematosus (SLE) pathogenesis, promoting B-cell response. IL10 is mainly secreted by regulatory T-cells, but follicular helper T-cells (T FH ), also produce it. We previously identified subset of CCR6 + IL-7R human tonsils providing IL-10-dependent help. These were able to IL-10, inducing IgG production. Objectives: investigate possible CD4 IL7R SLE pathogenesis. Methods: 37 patients fulfilling the ACR criteria for have been included....
