Fibrosis is mediated partly by extracellular matrix-depositing fibroblasts in the heart. Although these mesenchymal cells are reported to have multiple embryonic origins, functional consequence of this heterogeneity unknown. We sought validate a panel surface markers prospectively identify cardiac fibroblasts. We elucidated developmental origins and characterized their corresponding phenotypes. also determined proliferation rates each subset after pressure overload injury. We showed that...

Significance The nature of postnatal cardiomyogenesis in mammals remains dispute. Here, we provide cell-level evidence for the birth cardiomyocytes newborn and adult mice. Our clonal analysis, based on mosaic analysis with double markers mouse model, shows that are parent cell origin generated postnatally. findings confirm limited, symmetric division is a rare phenomenon heart after birth.

Abstract Aging, often considered a result of random cellular damage, can be accurately estimated using DNA methylation profiles, the foundation pan-tissue epigenetic clocks. Here, we demonstrate development universal pan-mammalian clocks, 11,754 arrays from our Mammalian Methylation Consortium, which encompass 59 tissue types across 185 mammalian species. These predictive models estimate age with high accuracy ( r > 0.96). Age deviations correlate human mortality risk, mouse somatotropic...

The pluripotent nature of human embryonic stem cells (hESCs) makes them convenient for deriving therapeutically relevant cells. Here we show using Wnt reporter hESC lines that the are heterogeneous with respect to endogenous signalling activity. Moreover, level activity in individual correlates differences clonogenic potential and lineage-specific differentiation propensity. addition protein or, conversely, a small-molecule inhibitor (IWP2) reduces heterogeneity, allowing stable expansion...

Ischemic heart disease is the leading cause of death worldwide. Myocardial infarction results in an irreversible loss cardiomyocytes with subsequent adverse remodeling and failure. Identifying new sources for promoting their formation represents a goal cardiac biology regenerative medicine. Within past decade, many types putative stem cells (CSCs) have been reported to regenerate injured myocardium by differentiating into cardiomyocytes. Some these CSCs translated from bench bed therapeutic...

Applications of embryonic stem cells (ESCs) require faithful chromatin changes during differentiation, but the fate X chromosome state in differentiating ESCs is unclear. Female human ESC lines either carry two active chromosomes (XaXa), an Xa and inactive with or without XIST RNA coating (XiXIST+Xa;XiXa), eroded Xi (XeXa) where no longer expresses has partially reactivated. Here, we established XiXa, XeXa, XaXa followed their differentiation. Surprisingly, found that pre-existing primed...

The cellular mechanisms driving cardiac tissue formation remain poorly understood, largely due to the structural and functional complexity of heart. It is unclear whether newly generated myocytes originate from stem/progenitor cells or pre-existing cardiomyocytes that re-enter cell cycle. Here, we identify source new during mouse development after injury. Our findings suggest progenitors maintain proliferative potential are main development; however, onset αMHC expression leads reduced...

ABSTRACT Aging is often perceived as a degenerative process resulting from random accrual of cellular damage over time. Despite this, age can be accurately estimated by epigenetic clocks based on DNA methylation profiles almost any tissue the body. Since such pan-tissue have been successfully developed for several different species, we hypothesized that one build pan-mammalian measure in all mammalian species. To address generated data using 11,754 arrays, each profiling up to 36 thousand...
