Abstract The presence of circulating tumor cell (CTC) clusters is associated with disease progression and reduced survival in a variety cancer types. In breast cancer, preclinical studies showed that inhibitors the Na + /K ATPase suppress CTC block metastasis. Here we conducted prospective, open-label, proof-of-concept study women metastatic where primary objective was to determine whether treatment inhibitor digoxin could reduce mean cluster size. An analysis nine patients treated daily...

Abstract Bone marrow haematopoietic stem cells (HSCs) are vital for lifelong maintenance of healthy haematopoiesis. In inbred mice housed in gnotobiotic facilities, the top hierarchy is occupied by dormant HSCs, which reversibly exit quiescence during stress. Whether HSC dormancy exists humans remains debatable. Here, using single-cell RNA sequencing, we show a continuous landscape highly purified human bone HSCs displaying varying degrees dormancy. We identify orphan receptor GPRC5C,...

Abstract Circulating tumor cells (CTCs) play a pivotal role in metastasis, the leading cause of cancer-associated death. Recent improvements CTC isolation tools, coupled with steady development multiomics technologies at single-cell resolution, have enabled an extensive exploration biology, unlocking insights into their molecular profiles. A detailed portrait requires interrogation across various levels encompassing genomic, epigenetic, transcriptomic, proteomic and metabolic features. Here,...

Leukemia stem cells (LSCs) share numerous features with healthy hematopoietic (HSCs). G-protein coupled receptor family C group 5 member (GPRC5C) is a regulator of HSC dormancy. However, GPRC5C functionality in acute myeloid leukemia (AML) yet to be determined. Within patient AML cohorts, high levels correlated poorer survival. Ectopic Gprc5c expression increased aggression through the activation NF-κB, which resulted an altered metabolic state intracellular branched-chain amino acids...

Glucosidations of oleanolic acid (1) and its dihydroxy-olide derivatives (2) were carried out to provide eight glycosides. All synthesized compounds evaluated by in vitro alpha-glucosidase inhibitory activity assay. 3-Acetyl derivative 15 showed the most potent effect. Structure-activity relationships within these are discussed.

In conventional analysis methods of soil slope, we must presume potential sliding surface at first and then gain the safety factors, judge stability slope by factors. fact, if know a is unstability but don’t where will be slide or failure, can’t adopt effective measures to support prevent accidents appearing. This paper use RFPA-Centrifuge numerical simulate failure with different without any surface, success process corresponding From calculate results RFPA-Centrifuge, can find point all...

A novel watershed algorithm using mountain areas simulation method to segment the field emission images was proposed for first time. In algorithm, sites in are compared many mountaintops. The small fluctuations mountains erased by filling method. And then traditional transform is applied get roughly. At last parameters of relative height needed a mountaintop used series image process dilation and comparation. This segmentation very easy be accomplished MATLAB have high efficiency. Results...

To well develop coal gas and prevent outburst accidents, pre-drainage project has become the prerequisite of mining in underground collieries. Its construction quality determined drainage effect as touched miners lives closely. Based on X-R s control chart, a novel tracking warning method for was proposed. Firstly, data structure upward cross-seam boreholes that were widely used built. As time went by, formed multivariable series. Using difference transformation between design, inspection...

The T 2 statistic is one important indicator of statistical process control theory to identify anomalies the multivariate industrial process. In research field coal gas pre-drainage control, previous achievements mainly based on univariate chart, which leaded huge workload and facilitated some human errors. Against these problems, a more comprehensive easy-to-use method was proposed. First at all, basic thought principle chart elaborated. Secondly, data structure samples were provided after...
