Epithelial-mesenchymal transition (EMT) is reported to involve in the crosstalk between tumor cells and tumor-associated macrophages (TAMs). Exosomes are considered as important mediators of orchestrating intercellular communication. However, underlying mechanisms by which EMT-colorectal cancer (CRC) promote M2 polarization TAMs remain less understood. In this study, we found that EMT-CRC promoted M2-like directly transferring exosomes macrophages, leading a significant increase...
Significance Breast cancer (BrCa) is the most common worldwide, and high-performance metabolic analysis emerging in diagnosis prognosis of BrCa. Here, we used nanoparticle-enhanced laser desorption/ionization mass spectrometry to record serum fingerprints BrCa seconds, achieving high reproducibility low consumption direct detection. Our analytical method, combined with aid machine learning algorithms, was demonstrated provide diagnostic efficiency accuracy 88.8% desirable prognostic...
New particles beyond the standard model of particle physics, such as axions, can be effectively searched through their interactions with electrons. We use large liquid xenon detector PandaX-4T to search for novel electronic recoil signals induced by solar neutrinos anomalous magnetic moment, axionlike particles, dark photons, and light fermionic matter. A detailed background is established using latest datasets <a:math xmlns:a="http://www.w3.org/1998/Math/MathML"...
Abstract The pattern recognition receptor, RAGE, has been shown to be involved in adaptive immune responses but its role on the components of these is not well understood. We have studied effects a small molecule inhibitor RAGE and deletion receptor (RAGE−/− mice) T cell autoimmunity allograft rejection. Syngeneic islet graft rejection was reduced NOD B6 mice treated with TTP488, (p < 0.001). RAGE−/− streptozotocin-induced diabetes showed delayed allografts compared wild type (WT)...
Accumulating evidence shows that, the dysregulation of circular RNAs (circRNAs) is associated with progression multiple malignancies. But, underlying mechanisms by which has_circ_0032627 (circDLST) contributed to gastric cancer (GC) remain undocumented. The expression and cellular localization circDLST its association clinicopathological characteristics prognosis in patients GC was analysed using fluorescence situ hybridization. Gain- loss-of-function experiments as well a subcutaneous...
MicroRNAs (miRNAs) can act as oncogenes or tumor suppressors by controlling cell proliferation, differentiation, metastasis and apoptosis, miRNA dysregulation is involved in the development of pancreatic cancer (PC). Our previous study demonstrated that Gabra3 plays critical roles progression. However, whether regulated miRNAs PC remains unknown. The expression levels miR-92b-3p were measured quantitative PCR (qPCR), immunoblotting, situ hybridization (ISH) immunohistochemistry (IHC)....
We report constraints on light dark matter through its interactions with shell electrons in the PandaX-II liquid xenon detector a total $46.9\text{ }\text{ }\mathrm{tonnes}/\mathrm{day}$ exposure. To effectively search for these very low energy electron recoils, ionization-only signals are selected from data. 1821 candidates identified within an ionization signal range between 50 and 75 photoelectrons, corresponding to mean electronic recoil 0.08 0.15 keV. The 90% C.L. exclusion limit...
Abstract Bile acids (BAs), well-defined signaling molecules with diverse metabolic functions, play important roles in cellular processes associated many cancers. As one of the most common BAs, deoxycholic acid (DCA) is originally synthesized liver, stored gallbladder, and processed gut. DCA plays crucial various tumors; however, functions molecular mechanisms gallbladder cancer (GBC) still remain poorly characterized. Here, we analyzed human GBC samples found that was significantly...
We report the first dark matter search results using commissioning data from PandaX-4T. Using a time projection chamber with 3.7 tonne of liquid xenon target and an exposure 0.63 tonne·year, 1058 candidate events are identified within approximate nuclear recoil energy window between 5 100 keV. No significant excess over background is observed. Our set stringent limit to matter-nucleon spin-independent interactions, lowest excluded cross section (90% C.L.) 3.8×10^{-47} cm^{2} at mass 40 GeV/c^{2}.
Clostridioides difficile is a pathogen contributing to increased morbidity and mortality of patients with inflammatory bowel disease (IBD). To determine how C. affects the severity colitis, we constructed dextran sulfate solution-induced colitis model challenged difficile. Without antibiotic administration, led transient colonization in mice but still significantly enhanced as assessed by weight loss, histopathological damages, cytokine concentrations. Because this effect independent toxin...
Timely screening of neuromyelitis optica spectrum disorder (NMOSD) and differential diagnosis from myelin oligodendrocyte glycoprotein associated (MOGAD) are the keys to improving quality life patients. Metabolic disturbance occurs with development NMOSD. Still, advanced tools required probe metabolic phenotype Here, we developed a fast nanoparticle-enhanced laser desorption/ionization mass spectrometry assay for multiplexing fingerprints (MFs) trace plasma cerebrospinal fluid (CSF) samples...
Abstract Overexpressed pro-survival B-cell lymphoma-2 (BCL-2) family proteins BCL-2 and BCL-X L can render tumor cells malignant. Leukemia drug venetoclax is currently the only approved selective inhibitor. However, its application has led to an emergence of resistant mutations, calling for drugs with innovative mechanism action. Herein we present cyclic peptides (CPs) nanomolar-level binding affinities or , further reveal structural functional mechanisms how these CPs target two in a...
Axionlike particles (ALPs) and dark photons (DPs) are viable matter particle candidates. We have searched for possible ALP/DP signals in the PandaX-4T liquid xenon detector using 440 kg·yr of data. A binned likelihood fit is constructed to search mono-energetic peaks induced by absorption processes between ALPs/DPs atomic electrons xenon. detailed temporal model decays associated with isotopes introduced constrain number background events. No signal excess over expectations observed, we...
Exosomes are 30-150 nm small membrane vesicles that released into the extracellular medium via cells function as a mode of intercellular communication. Dendritic cell (DC)-derived exosomes modulate immune responses and prevent development autoimmune diseases. Moreover, Schistosoma japonicum eggs show modulatory effects in mouse model colitis. Therefore, we hypothesized derived from DCs treated with S. soluble antigen (SEA; SEA-treated DC exosomes) would be useful for treating inflammatory...
Background: Circulating exosomal microRNAs (miRNAs) are considered as potentially non-invasive biomarkers for early detection and prognosis of cancers. Due to the lack highly sensitive specific molecular markers, a lot patients with hepatocellular carcinoma diagnosed in advanced stages. This study aims explore expression mode clinical value serum miR-34a HCC, providing new potential targets theoretical basis diagnosis monitoring carcinoma. Methods: The 60 HCC before after operation healthy...
Rapid and accurate clinical staging of pediatric patients with inflammatory bowel disease (IBD) is crucial to determine the appropriate therapeutic approach. This study aimed identify effective, convenient biomarkers for IBD in patients. We recruited cohorts varying severities compare features intestinal microbiota metabolites between active remitting stages. Metabolites potential were targeted further assessment both colitis model mice. The performance these markers was determined using...
