Despite NUDT15 variants showing significant association with thiopurine-induced adverse events (AEs) in Asians, it remains unclear which of or whether additional genetic should be tested to predict AEs. To clarify the best pharmacogenetic test used clinically, we performed studies and haplotypes AEs, genome-wide study (GWAS) discover variants, ROC analysis select model severe Overall, 2630 patients inflammatory bowel disease (IBD) were enrolled genotyped for codon 139; 1291 treated...

Cyclosporine is a potent immunomodulator and has beneficial effect in the treatment of ulcerative colitis (UC). We analyzed mechanism effects cyclosporine on regulation epithelial apoptosis via TGF-beta-related signaling, because balance between regeneration cells seems to be key factor maintain intestinal homeostasis. For this purpose, was induced by 4% dextran sulfate sodium (DSS), with anti-TGF-beta antibody assessed. Treatment ameliorated body weight loss, mucosal destruction,...

Background: Maintenance of mucosal healing may lead to a better outcome in patients with Crohn's disease (CD). Magnetic resonance diffusion-weighted imaging (MR-DWI) scans 1 year after infliximab (IFX) induction therapy were assessed as predictors maintained response, or remission, through 3 years treatment CD. Summary: MR-DWI and endoscopy data prospectively collected throughout IFX treatment. Altogether, 86 lesions from 13 given (weeks 0, 2 6) maintenance (5 mg/kg every 8 weeks beginning...

Granulocyte and monocyte adsorptive apheresis (GMA) has shown efficacy in patients with active Crohn's disease (CD). However, routine weekly therapy, it may take several weeks to achieve remission. This study was performed assess clinical safety of intensive GMA CD. In an open-label, prospective, randomized multicentre setting, 104 CD activity index (CDAI) 200 450 received GMA, at two sessions per week (n = 55) or one session 49). Clinical remission defined as a CDAI score <150. Patients...

Summary Toll-like receptor 4 (TLR4), which recognizes lipopolysaccharides, plays an important role in the innate immune response. In this study, we investigated of TLR4 development experimental colitis with regard to biological actions macrophage migration inhibitory factor (MIF) using null (–/–) mice. TLR4–/– mice were given 2% dextran sulphate sodium (DSS) drinking water induce colitis, was clinically and histologically as severe that seen wild-type (WT) The level tumour necrosis (TNF)-α...

Abstract Background Dietary fatty acids can affect chronic intestinal inflammation and have been reported to be associated with the development of ulcerative colitis (UC), mainly in Europe United States. The association dietary intake risk for UC was investigated Japan, where habits lead lower meat higher fish consumption than Western countries. Methods A multicenter case-control study 83 newly diagnosed patients 128 age- sex-matched control hospital conducted from 2008 2014. acid preceding...

We investigated the effect of p60, a virulence factor Listeria monocytogenes, on host immune response in vitro and vivo. Administration p60 before sublethal infection with L. monocytogenes enhanced innate resistance naïve mice. Mouse macrophage RAW264.7 cells produced tumor necrosis (TNF)-alpha to stimulation recombinant p60. Toll-like receptor 4 may be involved TNF-alpha production from induced by administration. Our findings demonstrated that modulates responses against infection.

Abstract Background and Aims This post hoc analysis assessed the efficacy safety of upadacitinib in patients with moderately to severely active ulcerative colitis stratified by corticosteroid use from Phase 3 clinical trial programme. Methods Patients were randomised [1:2] 8 weeks’ placebo or 45 mg once daily; Week responders re-randomised [1:1:1] 52 15 30 daily. Corticosteroid dose was kept stable during induction but tapered according a protocol-defined schedule [or investigator...

Ulcerative colitis (UC) patients harbor activated myeloid leukocytes, which exacerbate and perpetuate UC by releasing inflammatory cytokines. Granulocyte monocyte adsorptive apheresis (GMA) with an Adacolumn depletes elevated inducing efficacy favorable safety. To understand how the clinical outcome GMA is affected prior corticosteroid treatment or concomitant immunomodulators, a retrospective multicenter study in 102 patients, who had not responded well to first-line medications was...

Inflammatory bowel diseases [IBDs], including Crohn's disease [CD] ulcerative colitis [UC], are associated with mCAs, patients at malignancy development and/or modification of IBD phenotypes. In present study, we screened presence mCAs explored possible pathophysiological genetic mCAs. We analysed in peripheral blood from 3339 investigated clinical CD exposure to...
