Basic life science literature is rich with information, however methodically quantitative attempts to organize this information are rare. Unlike clinical research, where consolidation efforts facilitated by systematic review and meta-analysis, the basic sciences seldom use such rigorous methods. The goal of study present a brief theoretical foundation, computational resources workflow outline along working example for performing or rapid reviews research followed meta-analysis. Glioblastoma (GBM) patients suffer from a dismal prognosis, with standard of care therapy inevitably leading to therapy-resistant recurrent tumors. The presence cancer stem cells (CSCs) drives the extensive heterogeneity seen in GBM, prompting need for novel therapies specifically targeting this subset tumor-driving cells. Here, we identify CD70 as potential therapeutic target GBM CSCs.

Bone metastasis from breast and prostate carcinomas is facilitated by activation of bone-resorbing osteoclasts. Using proteomics approaches, we have identified peroxiredoxin-4 (PRDX4) as a cancer-secreted mediator osteoclastogenesis. We now report characterization L-plastin in the conditioned media (CM) MDA-MB-231 human cancer cells using immunoblotting mass spectrometry. The osteoclastogenic potential CM with siRNA-silenced was significantly reduced. detected cancer-derived exosomes,...

Abstract Background Resolving the differential diagnosis between brain metastases (BM), glioblastomas (GBM), and central nervous system lymphomas (CNSL) is an important dilemma for clinical management of main three intra-axial tumor types. Currently, treatment decisions require invasive diagnostic surgical biopsies that carry risks morbidity. This study aimed to utilize methylomes from cerebrospinal fluid (CSF), a biofluid proximal tumors, reliable non-invasive classification addresses...

Abstract Osteogenesis imperfecta (OI) is a rare genetic disorder commonly caused by variants of the type I collagen genes COL1A1 and COL1A2. OI associated with increased bone fragility, deformities, pain, reduced growth. Setrusumab, neutralizing antibody to sclerostin, areal mineral density (aBMD) in 21-week phase 2a dose escalation study. The 2b Asteroid (NCT03118570) study evaluated efficacy safety setrusumab adults. Adults clinical diagnosis I, III, or IV, pathogenic variant COL1A1/A2,...

During gastrulation, Wnt-β-catenin signaling dictates lineage bifurcation generating different mesoderm cell types. However, the specific role of Wnt receptors in specification remains elusive. Using selective Frizzled (FZD) and LRP5/6 antibody-based agonists, we examined FZD receptors' function during directed differentiation human pluripotent stem cells (hPSCs). We found that FZD2 FZD7 are expressed at membrane hPSCs their activation triggers β-catenin with kinetics, thereby influencing...

Abstract Glioblastoma (GBM) remains a formidable challenge in neuro-oncology, characterized by heterogeneity and aggressive tumor growth. The identification of novel biomarkers therapeutic targets is crucial for advancing promising therapies. This study profiled patient-derived samples utilizing single-cell RNA sequencing proteomics platforms to uncover the upregulation glycoprotein non-metastatic melanoma protein B (GPNMB) cells, particularly post-treatment recurrent as well...

Abstract Background Central nervous system lymphoma (CNSL), is a rare subtype of non-Hodgkin lymphoma, primarily affecting the brain and spinal cord. Most therapeutic systemic agents have limited penetration blood-brain blood-cerebrospinal fluid barrier, with latter potentially promoting treatment “sanctuary” for cancer cells. Evaluation occult disease, particularly in cerebrospinal (CSF), challenging. In clinical experience addition multi-agent intraventricular chemotherapy (MAIVC),...

Bone cells sense and actively adapt to physical perturbations prevent critical damage. ATP release is among the earliest cellular responses mechanical stimulation. Mechanical stimulation of a single murine osteoblast led 70 ± 24 amole ATP, which stimulated calcium in neighboring cells. Osteoblasts contained ATP-rich vesicles that were released upon Surprisingly, interventions promoted vesicular reduced release, while inhibitors potentiated release. Searching for an alternative route, we...

Single-cell RNA-sequencing (scRNA-seq) offers functional insight into complex biology, allowing for the interrogation of cellular populations and gene expression programs at single-cell resolution. Here, we introduce scPipeline, a data analysis toolbox that builds on existing methods modular workflows multi-level annotation user-friendly reports. Advances to scRNA-seq include: (i) co-dependency index (CDI)-based differential expression, (ii) cluster resolution optimization using...

The P2 purinergic receptor family implicated in many physiological processes, including neurotransmission, mechanical adaptation and inflammation, consists of ATP-gated non-specific cation channels P2XRs G-protein coupled receptors P2YRs. Different cells, bone forming osteoblasts, express multiple receptors; however, how P2X P2Y interact generating cellular responses to various doses [ATP] remains poorly understood. Diagnosis and prognostication of intra-axial brain tumors hinges on invasive sampling, which carries risk morbidity. Minimally-invasive sampling proximal fluids, also known as liquid biopsy, can mitigate this risk.
