Recessive or dominant inborn errors of type I interferon (IFN) immunity can underlie critical COVID-19 pneumonia in unvaccinated adults. The risk children, which is much lower than adults, remains unexplained. In an international cohort 112 children (<16 yr old) hospitalized for pneumonia, we report 12 (10.7%) aged 1.5–13 with (7 children), severe (3), and moderate (2) 4 the 15 known clinically recessive biochemically complete IFN immunity: X-linked TLR7 deficiency children) autosomal...

Patients with autoimmune polyendocrinopathy syndrome type 1 (APS-1) caused by autosomal recessive AIRE deficiency produce autoantibodies that neutralize I interferons (IFNs)1,2, conferring a predisposition to life-threatening COVID-19 pneumonia3. Here we report patients NIK or RELB deficiency, specific of autosomal-dominant NF-κB2 also have neutralizing against IFNs and are at higher risk getting pneumonia. In these found only in individuals who heterozygous for variants associated both...

We found that 19 (10.4%) of 183 unvaccinated children hospitalized for COVID-19 pneumonia had autoantibodies (auto-Abs) neutralizing type I IFNs (IFN-α2 in 10 patients: IFN-α2 only three, plus IFN-ω five, and IFN-α2, IFN-β two; nine patients). Seven (3.8%) Abs at least ng/ml one IFN, whereas the other 12 (6.6%) 100 pg/ml. The auto-Abs neutralized both unglycosylated glycosylated IFNs. also detected pg/ml 4 2,267 uninfected (0.2%) 45 (2%). odds ratios (ORs) life-threatening were, therefore,...

We compared whole-exome sequencing (WES) GWSA for this purpose. analyzed 110 subjects originating from different regions of the world, including North Africa Middle East, which poorly covered by public databases have high consanguinity rates. tested applied a number quality control (QC) filters. Methods: A retrospective was conducted by pediatric infectious specialists from 32 different hospitals all over Turkey case record forms. Pediatric cases who were diagnosed as COVID-19 between March 16, 2020, June 15, 2020 included. Case characteristics including age, sex, dates onset diagnosis, family, contact information recorded. Immunoprophylaxis is routinely recommended for infants born to mothers with hepatitis B virus (HBV) infection within the first 12-24 hours. Detection of surface antibody (HBsAb) resulting from immunoglobulin administered at birth may be perceived as a real vaccine response. This makes it difficult detect HBV infection. For this reason, that antigen positive and who received immunoprophylaxis should have HBsAb testing when they are 9-15 months old.

Inborn errors of IFN-γ immunity can underlie tuberculosis (TB). We report three patients from two kindreds without EBV viremia or disease but with severe TB and inherited complete ITK deficiency, a condition associated that renders immunological studies challenging. They have CD4+ αβ T lymphocytopenia concomitant expansion CD4−CD8− double-negative (DN) Vδ2− γδ lymphocytes, both displaying unique CD38+CD45RA+T-bet+EOMES− phenotype. Itk-deficient mice recapitulated an the DN lymphocyte...
